The story of Despatch follows veteran crime journalist Joy, once the titan of the newsroom, who finds himself on the brink of obscurity in the modern era that's dominated by digital media. The teaser of ZEE5’s Despatch kicks off with a sudden stone pelting inside a house that terrifies a young man named Joy (Manoj Bajpayee), who is quietly having his meal. Soon he gets a call from an unknown number where the anonymous person threatens him that he must leave whatever he's trying to do.

In the next scene, we see a woman claiming that in the media industry, information is the only thing that's valued. Soon, we see another man saying that someone is involved in a Rs 8000 crore scam. This leads Joy to find out the person behind the scam by any means. He begins to meet several people and do his research.
